It can be turned on/off at any given time. It's also always disabled in your favorites, so that shouldn't really be a concern of "missing stuff because I blocked that girl/guy".
Populating the list and creating
custom queries that exclude all filters allows you to include those uploaders just in case and ignore them the rest of the time.
Bookmarking or "storing" those queries generally is a good idea as long as you don't end up bookmarking hundreds of similar or obsolete queries.
